Derek Lowe has had several posts on that upcoming problem on his blog, http://pipeline.corante.com/ , over the past 6 months or so. And I strongly recommend his blog to anyone interested in pharmacology. He spends more time on the business and politics aspects than I really like, but also has good posts on scientific method and chemistry, his "Things I Won't Work With" series, http://pipeline.corante.com/archives/things_i_wont_work_with/ , is particularly entertaining.
